I have been thinking of this for a couple of days now, it is very hard to choose just one game. Because like most of us here, i have playing games since the 80's.
I think i would go for the Ultima underworld. My friend had this game on his PC, and we did spend a lot of time playing this great game.

Hybrus was the reason I got an Amiga. then on the PC has to be Duke Nukem 3d. it was just all class

2. Worms Directors Cut on Amiga
3. SWOS on Amiga
4. Rainbow Six Vegas 2 on Xbox 360
5. SuperFrog on Amiga
6. Mortal Kombat Trilogy on Playstation One
7. WWF No Mercy on N64
8. Streets of Rage 2 on Megadrive
9. New Super Mario Bros on Nintendo DS

Amiga:  Speedball 2, Worms, Populous 2

PC:

Star Wars:  Knights of the Old Republic, 1 & 2
Half-Life 2 plus episodes
Starcraft, Starcraft 2
Tomb Raider 1, 2, and 4.
UFO Enemy Unknown (I've never seen it run well on the Amiga, plus the graphics were better on the PC).

N64:  Goldeneye.  Wipeout 64.

If I had to pick one out of all of these, KotoR 1 or 2 would be high on the list of possibilities.

It is difficult every plataform and system have their key game.

Yes this is indeed a tough question. How to narrow all my favourite games down to one, I guess it must be one per computer/console for me as well, so here goes:

Call of Duty (PC)
Tomb Raider (PS1)
Pinball Dreams (Amiga)
ThunderBlade (C64)

But ask me the same question in a year from now on, and the answer might be different.

While I understand the many people who have said this is a difficult question, for me it's easy:  Archon.  I first played this on the Atari 800 and then on the Amiga.  Recently I discovered that there's a decent version of Archon for the iPhone now!  Woohoo!
