If there is enough physical space, it should.

The Clockport Adapter for the 68k socket of your Amiga 500 (or Amiga 1000)...

I have one in my A500, and it fits nicely with a little modification to the shield. And I've got a little clockport cable coming out the side expansion slot. I haven't actually used it yet (working on it!), so I can't comment on functionality, unfortunately.
