I can confirm that only RED and BLUE button are working as intended while trying to play a game with CD32 controls

Just saw that is is a known issue at least for non CD32 games : https://forum.amiga.org/index.php?topic=76709.0

So, with something like Gloom, how would I use the GS controller and have the start button work as it would on the CD32?

Wait for Joypad remapping, not possible at the moment AFAIK
