I am having the exact same problem.  I have tried it with and without my trapdoor card and I have tried a multitude of cards with it (both CF and SD) and I cant get it working :-(

Using Blizkick to make Amiga 3.1 so it runs 3.9
PCMCIA CF reader = PCMCIA Adapter Compact Flash Type II
CF card = Kingston 2GB or Sandisk 1GB

Thats pretty much what I have tried too!  i.e. just about everything.  And yes, I have everything installed and get the "no disk" when I access CF0:  And, Prepcard sees a card - just nothing... :(

I have tried fat and fat32 - neither work :-(
