Check the icon properties. You'd need more specifics. For example, if it says "Unable to open SYS:Utilities/More" when your More program (a text viewer) is in SYS:C. Just an example. ;)

Hummm.  Have you been copying or changing icons?  I wonder if their icon type is not set correctly?

If a "Tool" icon got changed to a "Project" icon, for example.  But I'd still start by checking your default tools...

Yes.  It still could be multiple things, and SnoopDos would help, but I'd start by looking at what your icon properties are.

Post some screenshots of what you see when you look at the icon properties, especially the "default tool" section.

Aaah!  It appears you've changed the icon type of your Music: partition from a disk into a Tool.  :(

Right-click and get the properties for any of your other partitions (assuming any of the others do still work).  You should be able to see the difference.

Edit, hold on, I'm still having a hard time understanding your screenshot.  So you have a partition called "Music:", and inside that you have a program called "PT315", is that correct?  Do you have Dopus or any other file manager utility installed?  If not, drop down to the shell and get a directory listing of Music:.  You might be missing the file for PT315 - in other words, you have the .info icon file, but not the program itself.  Or the protection bits for it got changed.

Do this:  drop down to the shell and type this, and post a screenshot:

Side thought:  are you using a mechanical hard drive or a CF or similar?  Possible your hard drive might be beginning to fail?

Try dragging the PT315 icon into RAM: and running it from there.  Any difference?

For the heck of it, I just downloaded PT315 from Aminet.  Runs fine for me, but my goodness, is that a strange interface.  ;)

I was able to replicate your error by mucking up the protection bits on the file.  This can also happen accidentally (for example, when copying a file).  Please post a directory listing to verify if this is the issue you're having.  See attached screenshot!  :)

How is your drive partitioned, and what file system?  Are you getting file transfer errors or file corruption?  Or just that one error message?  I was under the assumption that everything was working well, except for that one error message?

Hate to say it, but yes, personally, I would.  Better safe to do it now than when you encounter more weird errors in a week or a month.  Thomas could probably give you a better answer but I haven't seen him on here in a little while...
