If you mean the 2 9-pin ports on the side of the machine, those are the mouse and joystick ports, not serial ports. You need them both, as I don't think mice usually work properly in the joystick port.
Ancient serial mice can be connected to the actual serial port with the help of a driver, but they don't work with non-system-friendly programs.
