First, I would upgrade to version 4.4 of Poseidon, and use the z3 device driver (as you have an A3000), and then check to make sure Trident saved your settings.  Make sure the contacts are all cleaned on the Zorro connector and at the USB sports failing that, empty the onboard memory and reset the card to its native state.

I booted up one of my A4000D's and changed the CD file system, hit the Save Default button, and it remained changed through every boot.

You can also load your USB stack with the Startup-Sequence command:
Prefs/Trident -NOGUI
You may need a Run command in front

Classes like that are stored in the Classes directory on your SYS: partition.  Until you get it working properly, I would empty or disable the ROM.
