There have been a lot of great Amiga games, and a we have lot of threads about great memories of those.
How about a thread that captures the best laugh you've had playing a game? That possibly unexpected moment of pure hilarity that inspires running jokes that may have survived the 15 or 20 years now since they happened. There anything you fire up once in a while that still brings side-paining laughter fits to everyone in the room?
For me, one of the classics has to be Big Business. On the surface, it tries to be a fairly straight-forward manufacturing business management game. You have budgets to balance, resource inventories to manage, manufacturing plants to build and set productions on, etc. In MULE fashion, there are resource auctions every round, and other standard game mechanics.
Now, the game is intentionally lighthearted. There are plenty of Easter Eggs to find by clicking around on the screen. (For example, you can shoot the test rat in the R&D lab, and occasionally you'll even get sued for it.) You can also plot some industrial espionage against your fellow players. If your company is succeeding, the board room graphics get updated, if your company is failing, the marketing director starts showing up drunk, etc. And for the first few rounds everything stays pretty much on the tracks.
But then the insanity and true hilarity starts creeping in. A minor purchase transaction may send the boardroom from champagne glasses to empty beer cans. That small loan stacked up into billions of dollars, overnight. Some bidding in the auctions goes haywire -- someone paid you to take their inventory! Of course your workers went on strike, but that actually turned out to be good fortune, as one of the raw materials you were paid to take last turn is now worth more than you were hoping to sell the finished goods for. Among the chaos the headlines start getting more bizarre, players start getting multi-billion dollar loans without even applying for them, and someone just got sued for $4, but had to pay millions in court fees. And finally one of the companies gets bailed out to the tune of a few trillion dollars, just before the game culminates with a Guru Meditation.
A truer picture of the complexities of economy may never be painted again. We tried a drinking game over this game once, but no one has enough memory of it to reassemble the rules.

Oh yeah, many a comedic game to be played on the Amiga. I never had a chance to play Big Business, but that sounds awesome - if not a little too realistic and sad on levels. For me, games like Keef the Thief, Monty Python's Flying Circus(although some of it is just plain weird) and especially Leisure Suit Larry had me rolling back in the day. Leisure Suit Larry still does. It's cheesy, but as an adult, I can still appreciate (maybe more so now) what it's about.
I can't remember the name of it now(it's kind of a kooky/cartoony medieval theme), but there's a game with an intro or title screen where there's sounds of swords clanging, people "dying" and funny bits of speech like "no, no, no!" that's still pretty funny today. Same with the Cannon Fodder series. And before you think I'm uber grim or something, it's only funny because they're drawn/animated unrealistically and the sound bytes are hilarious. If Cannon Fodder were to be redone to look, behave and sound like real troops, I wouldn't care for it at all.
Now if that new AvP game has the Predator throwing his disc and decapitating someone (alien or human), I'd think that was pretty funny. lol

I think Police quest was pretty funny...
When you observing the drug deal in the park and you shoot the guy it says,
"You just blew that guy into Police quest 2"
Space quest 2 has the tasmanian devil, an Alien that french kisses you, and Vohaul squashes you with his fist after you're shrunken.

Wizkid ...... All of it, but especially one the first level where u flush the toilet (at the bottom of the well) and flood the well (yess the well), you know.... as you do.
LOL Classic

Years ago I got my A1200. I bought some magazines with coverdisks.
I played a game called Top Secret and I press for fun on the F10 key and it skips the level. Later I read in a magazine about it.
I tried to find a cheat for the playable demo Apocalypse and I pressed the left mouse button and the game crashed. later I played a Super Frog Demo an did the same and I found that the Superfrog becomes invisible.

I remember playing a game of ArcticFox, back when the game and my A1000 were pretty new. The game had an experimental (debug) mode in which you could make the tank pretty much do anything, including jump into the air.
One day, being the mischievous and evil soul that I am, I was playing the game in experimental mode and decided to launch a missile and activate the missile camera. While doing that, I made the tank jump into the air and tried to direct the missile into it. It was quite difficult to do, but in the effort of twisting and turning the missile so that I could smack into the tanks undercarriage, a upside-down smiley-faced alien (complete with antennae) appeared from the top of the view-port. Not funny, but it amused me greatly.
Never was able to recreate that one, though...

Worms was full of comedic moments. One time, I managed to get one to bounce of several mines before falling headlong into the water :-)

TD2, the Crying men image in Stunts Track Racer, the sliding player with rainy match in Microprose Soccer, Somethings in Ugh!, anothers things in Little Computer People, just for say somthings i remember in this moment..
But, in general, the Amiga Games are full with fun things, and thats give to you a funniest moments!.. Thats moments you still remember today!..

Funniest moments, lots of the above already posted and...
Mostly vocals based samples as well:
'Ergonomically terrific', 'We advanced a tech-level', 'ooo I don't like that!' and from memory 'The desert, unchanged for millions of years, yet witnessed to a biblical prophecy come true that one day the meek shall inherit the earth.'
Hmm what else is there...
'War has never been so much fun...' am sure you all know that one, god even further back 'hey hey its zoot' on the speccy 48k. There was this animation in the demo scene where 3 bright yellow chickens were beatboxing to some tune that was funny back in the day. Everything about Monkey Island I and II was great. The muppet song in Jesus on Es, Chuck Rock intro tune... love it. Who can't forget Amy the Squirrel?
Am sure there is more...

I accidently pressed the second button on my cd32 pad playing basejumpers last week. I thought the result was pretty funny, have a look:

(mind, the second button cannot be used in the game, its just a joke!)
also, monkey island 2 and gobliiins were quite hilarious at times.

No one has mentioned the Monkey Island games :(
Those games had me laughing for days, now and them when I play them from start to finnish I still laugh at them :D

North and South was full of funny bits.
The cartoons
The sounds
The Tickling the camera mans bum when setting up a game

This one looked like fun, but I didn't get very far not understanding German. http://www.mobygames.com/game/amiga/biing-sex-intrigue-and-scalpels

One of the funniest Amiga games vids I've ever seen:

I mean...I saw a lot of weird curiosities in SWOS 96/97, but this one is the best IMO :D

The cheat codes in Alien Breed are pretty funny.
"ST mode" always cracks me up
-
For me it's Moonstone. The beast that grabs the knight and squeezes it until the head pops out. The swamp creature making me jump out of my chair when it suddenly emerged from the mud. And the laugh I had playing with other 3 friends in same match. It's priceless.

Oh yeah, Base Jumpers was an excellent multi-player game for its unpredictable bloody deaths and truly ridiculous sub-games.
Here are a few of my comedic picks from disks gone by:
- Wings of Fury, for the sound the little Japanese soldiers made when machine gunned!
- Worms, for multiplayer laugh-at-your-mates oppurtunities and ridiculous weapons
- Putty, for the insane awards like "mashed banana and jam in a plastic bag for extreme heroism" and "herring cranium for bravery".
- Cannon Fodder, for the entertaining levels, music and level names. And the mass of gravestones on the hillside as your troops were slaughtered.
- Sensible Golf - terrible gameplay but I loved the sarcastic clapping when you missed a putt
- Super Skidmarks, for the name! And for allowing you to race against your mates as a cow on wheels
- Sensible Trainspotting (Amiga Power coverdisk) for being an actual train spotting simulation/parody. Classic!

The Monkey Island games had me in stitches. Those obscure puzzles like the spitting contest and the monkey wrench.
